Abstract

The synthesis of chiral compounds required as precursors of multiple synthesis chains in the pharmaceutical, food, and fragrance industries can be carried out by means of chemical processes or biocatalysis. Biocatalysis, in turn, encompasses two principally different processes: cell-free biocatalysis and whole-cell biotransformation. Whole-cell biotransformation has disadvantages with regard to yield and downstream processing. However, it is considered to be potentially more environmentally benign than chemical synthesis and more costeffective than cell-free biocatalysis. In the study presented here, ethyl-3-hydroxybutyrate (E3HB) was produced by whole-cell biocatalytic reduction. As part of the study, the fed-batch process was evaluated, both under aerobic and anaerobic culture conditions. The results have shown that the anaerobic processes produced yields YE3BH/EAA of 74%, compared to 88% that can be achieved in aerobic processes. The final maximum concentration of EAA was 20 gl-1 under aerobic and under anaerobic conditions. The gained CO2 and ethanol from the anaerobic process are byproducts that can be used in a biorefinery. Additionally, the process data were compared to a simulation of the processes that showed qualitatively good correspondence with the measured data courses. Quantitatively, it was possible to achieve an R² (coefficient of determination) between 0.99 and 0.29 for the formation of E3HB. The R² for EAA varied from 0.98 to -97.3. In this respect, the model must be improved further. The model was applied as a helpful tool for model-based data analysis of trial results.
