This multicenter study aims to establish a cohesive clinical phenotype. METHODS: Retrospective clinical data from 18 centers across 12 countries yielded 360 FTD patients with predominant RATL atrophy through initial neuroimaging assessments. RESULTS: Common symptoms included mental rigidity/preoccupations (78%), disinhibition/socially inappropriate behavior (74%), naming/word-finding difficulties (70%), memory deficits (67%), apathy (65%), loss of empathy (65%), and face-recognition deficits (60%). Real-life examples unveiled impairments regarding landmarks, smells, sounds, tastes, and bodily sensations (74%). Cognitive test scores indicated deficits in emotion, people, social interactions, and visual semantics however, lacked objective assessments for mental rigidity and preoccupations. DISCUSSION: This study cumulates the largest RATL cohort unveiling unique RATL symptoms subdued in prior diagnostic guidelines. Our novel approach, combining real-life examples with cognitive tests, offers clinicians a comprehensive toolkit for managing these patients. Highlights: This project is the first international collaboration and largest reported cohort. Further efforts are warranted for precise nomenclature reflecting neural mechanisms. Our results will serve as a clinical guideline for early and accurate diagnoses. date: 2024-08 date_type: published publisher: WILEY official_url: https://doi.org/10.1002/alz.14076 oa_status: green full_text_type: pub language: eng primo: open primo_central: open_green verified: verified_manual elements_id: 2296543 doi: 10.1002/alz.14076 medium: Print-Electronic lyricists_name: Warren, Jason lyricists_name: Fox, Nicholas lyricists_name: Rohrer, Jonathan lyricists_name: Barkhof, Frederik lyricists_id: JDWAR75 lyricists_id: NCIFO25 lyricists_id: JDROH34 lyricists_id: FBARK32 actors_name: Barkhof, Frederik actors_id: FBARK32 actors_role: owner funding_acknowledgements: AACSF-22-849085 [Alzheimer's Association]; R01-AG37491 [Josephs and Whitwell]; R01-DC010367 [Josephs and Whitwell]; R21-NS94684 [Josephs and Whitwell]; IDN/21/010 [KU Leuven]; C24/18/095 [KU Leuven]; [Sequoia Fund]; 2022-Projekt0080 [Swedish Federal Government]; [Fondation Planiol]; [Fondation France Alzheimer]; [CNPq]; K23 AG063900-01A1 [Pressman]; 2022-00775 [NIHR UCLH Biomedical Research Centre]; [Swedish Research Council]; 2017-0383 [Rankin]; [Knut and Alice Wallenberg Foundation]; [National Brain Appeal]; [Lund University]; AF-980907 [Swedish Alzheimer Foundation]; [Alzheimer's Society]; [Alzheimer's Research UK]; [Royal National Institute for Deaf People]; RF1-AG029577 full_text_status: public publication: Alzheimer's & Dementia: The Journal of the Alzheimer's Association volume: 20 number: 8 pagerange: 5647-5661 pages: 15 event_location: United States issn: 1552-5260 citation: Ulugut, Hulya; Bertoux, Maxime; Younes, Kyan; Montembeault, Maxime; Fumagalli, Giorgio G; Samanci, Bedia; Illan-Gala, Ignacio; ...
